Biological consequences of gain-of-function thyrotropin receptor (TSHR) mutant expression in adipose tissue

Baker G , Gregory J , Bakhsh A , Betts P , Hughes I , Ludgate M

Neonates harbouring germline gain-of-function TSHR mutations (M453T, L629F) have been reported with transient proptosis. This resolves once euthyroidism is achieved. In contrast children expressing TSHR mutations (V597L, I486T) fail to thrive, with weight gain and height between the 0.4th and 2nd centiles. We have previously demonstrated TSHR expression in preadipocytes undergoing adipogenesis. We now investigate the consequences of constitutively active TSHR mutation expressi...

Adverse metabolic correlations relate to free T3 levels in subclinical hypothyroidism; common FOXE1 polymorphisms associate with blood pressure

Introduction: The effects of mild subclinical hypothyroidism (SH; TSH <10 mU/l) on metabolic outcomes are unclear. This may relate to differences in aetiology, including thyroid autoimmunity or genetic factors such as TSH-receptor mutations (TSHR-M) and FOXE1 polyalanine tract length (FOXE1–PTL) polymorphisms, which are associated with altered thyroid function. We hypothesised that the metabolic manifestations of SH may depend upon its aetiology.<p class="abstext"...

Introduction: BC patients have a high prevalence of anti-TPO-autoantibodies (TPOAb), which seem to be protective in BC patients. We hypothesized the presence of T-lymphocytes citotoxicity against a common antigen between thyroid and BC cells.Aim: To evaluate the possible expression of TPO gene in BC.Materials: Frozen tissue specimens: 8 BC, 5 peri-tumoral breast tissues (PT), 3 pancreatic adenocarcinoma (P), 2 kidney cancers (K) an...
